基于肠道菌群和代谢组学探讨复方蜘蛛香凝胶贴膏治疗消化不良的作用机制

摘要:

目的:通过肠道菌群和代谢组学技术探讨复方蜘蛛香凝胶贴膏治疗消化不良的作用机制。方法:复制高热量饮食诱导的幼鼠消化不良模型(high-calorie-diet-induced dyspepsia, HC-DID),根据各组幼鼠体质量和摄食量变化、胃排空率、小肠推进率、胃窦和十二指肠病理变化评价复方蜘蛛香凝胶贴膏治疗消化不良的作用;ELISA法检测幼鼠血清中D-木糖、胆囊收缩素(CCK)、血管活性肠肽(VIP)、5-羟色胺(5-HT)和胃动素(MTL)含量;通过16S rDNA高通量测序和超高效液相色谱-四极飞行时间质谱法(UPLC-QTOF-MS)检测幼鼠肠道菌群和肠道内容物代谢物的变化。结果:与正常对照组相比,模型对照组幼鼠的摄食量和体质量明显降低,胃排空率和小肠推进率明显降低,血清D-木糖、MTL含量降低,CCK、VIP、5-HT含量升高(P<0.05或P<0.01);与模型对照组相比,复方蜘蛛香凝胶贴膏4.1、8.2、16.4 g/kg组消化不良幼鼠的摄食量和体质量增加,胃排空率和小肠推进率增加,血清D-木糖、MTL含量升高,CCK、VIP、5-HT含量降低(P<0.05或P<0.01)。复方蜘蛛香凝胶贴膏8.2 g/kg组能够增加消化不良幼鼠的肠道菌群多样性。幼鼠肠道内容物中鉴定出30种差异代谢物,精氨酸和脯氨酸代谢等5条通路是复方蜘蛛香凝胶贴膏干预后差异最显著的代谢通路。结论:复方蜘蛛香凝胶贴膏可以提高消化不良幼鼠的摄食量,促进胃肠动力,其机制可能与调节肠道菌群结构和精氨酸、脯氨酸代谢密切相关。

Abstract:

Objective:To explore the mechanism of Compound Zhizhuxiang(蜘蛛香) Gel Plaster in the treatment of dyspepsia based on gut microbiota and metabolomics techniques. Methods:A high-calorie-diet-induced dyspepsia(HC-DID) model was established in young mice. The therapeutic effect of Compound Zhizhuxiang Gel Plaster on dyspepsia was evaluated based on changes in body weight and food intake, gastric emptying rate, small intestinal propulsion rate, and pathological changes in the gastric antrum and duodenum. ELISA was used to detect the serum levels of D-xylose, cholecystokinin(CCK),vasoactive intestinal peptide(VIP),5-hydroxytryptamine(5-HT),and motilin(MTL). Changes in gut microbiota and metabolites in intestinal contents were analyzed by 16S rDNA high-throughput sequencing and ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-QTOF-MS). Results:Compared with the normal control group, the model control group showed significantly reduced food intake and body weight, as well as suppressed gastric emptying rate and small intestinal propulsion rate(P<0.05). Compared with the model control group, Compound Zhizhuxiang Gel Plaster significantly increased food intake and body weight, enhanced gastric emptying and small intestinal propulsion rates, and promoted gastrointestinal motility in dyspeptic mice(P<0.05). Moreover, Compound Zhizhuxiang Gel Plaster increased the diversity of gut microbiota in dyspeptic mice. A total of 30 differential metabolites were identified in the intestinal contents of young mice, and Compound Zhizhuxiang Gel Plaster could restore metabolic disturbances. Five metabolic pathways, including arginine and proline metabolism, were identified as the most significantly altered after intervention. Conclusion:Compound Zhizhuxiang Gel Plaster can improve food intake and promote gastrointestinal motility in dyspeptic young mice. Its mechanism may be closely related to the regulation of gut microbiota composition and arginine and proline metabolism.
